How much will all this cost and when can I get one?

Lexus isn't saying, and the company isn't even giving estimates yet. Our guess would be somewhere around the $35,000-$40,000 mark, considering that the least-expensive Lexus so far is the IS 250, and the HS has considerably more equipment in a similarly sized package. The 2010 HS 250h will go on sale in August, and prices are likely to be announced well before then.
